RESEARCH INTEREST AND ACHIEVEMENT

Dr Rong’s research has been focusing on the molecular characterization of anti-inflammatory, neuroprotective and neurotrophic activities of Chinese medicines towards the development of novel therapies for neurological and cardiovascular diseases. Our research is directed to three categories: 1) Molecular characterization of the cellular responses to complex herbal medicines by integrative genomic, proteomic, cell signaling and chemical biology approaches. Our strategy is to profile the biological responses to complex herbal medicine formulations by RNA-sequencing, DNA microarray and LC/MS-based proteomics. The potential genes or protein targets are selected from the transcriptomic or proteomic profiles in a bias-free manner. 2) Basic mechanisms underlying the neuroprotective, neurorestorative and cardioprotective activities of the bioactive botanicals. Our strategy is to identify the corresponding active compounds from the complex herbal medicine formulations through a bioactivity-guided fractionation procedure against specific disease-relevant molecular targets. 3) Structure-activity relationship of the bioactive botanicals and drug discovery. Our strategy is to develop more potent drug candidates by chemical biology approaches and molecular docking techniques. In the past a few years, we have selected a group of signature genes from transcriptomic and proteomic profiles as new therapeutic targets and quality control metrics. We strive to dissect the interactions between the active compounds in complex herbal medicines and the protein targets for the development of new chemically defined and mechanism-specific medicines. With the support of several GRF, HMRF and NSFC grants, we have successfully discovered the active compounds for the regulation of heme oxygenase-1 (HO-1), NADP-dependent leukotriene B4 12-hydroxydehydrogenase (LTB4DH), arginase-2, calreticulin, GRP78 and PKM2. The experimental results have been published as more than 60 papers and four international patents.
